"On the Road Again" by Willie Nelson (from the album Honeysuckle Rose, 1980)
This is track 1 on a mixed CD I made for a big road trip last summer. The CD is a melange of some real roadie tunes, including "Theme from Speed Racer" and Clarence "Frogman" Henry's "Ain't Got a Home." Made sure to put it at the top of the CD pile for when the car pulls out of the driveway sometime in late morning.

Where are you going again?
All over! Through the US and parts of Canada. I'll do a tabulation of how many states and provinces I touch through along the way.
I love that song.
Hehehehe. It makes me giggle, for unfathomable reasons.
It's one of the all-time great giggly sing-a-long songs. Hence it's roadtrip perfection.
Every time we left on a road trip, and I do mean every time, my dad would play that song as we pulled out of the driveway. My mom would roll her eyes, but I loved it. It was often followed, in those early heady days of the 1980's, by Kenny Rodger's "The Gambler", but still. Good times.
Just as it's good to be making music with your friends, you also got to know when to walk away, or when to run.
